Abstract:
Molecular studies play a considerable role in the modern world. In areas such as
Medicine, Agriculture, Animal husbandry, and even Forensic studies. DNA is the
fundamental material of the Molecular study. The first step of all Molecular studies
is extraction of DNA. DNA is stored in the nucleus, mitochondria or cytoplasm in
all living cells. The amount of DNA in the cells are vary with different organism.
DNA could be extracted from different body parts of the organism. Drop of blood,
hair, nails, skin, fins bones, and muscles are used for DNA extraction. Muscles are
the commonly used body part for DNA extraction from fishes. But muscle biopsy
is very harmful for fishes that it could lead to infection or affect the survival. This
situation become worse when small fishes specially the endangered and endemic
are concerned because it difficult to get a muscle sample without sacrificing the
small fishes.
Large numbers of DNA extraction protocols are used to extract DNA from
different body parts of organisms. The aim of the present study was to determine
the most suitable body part that exerts harm to the small fish species. To different
protocols were compared using different body parts.
In our study we observed that removal of a part of caudal fin is less harmful for'
survival of fish. It is the fastest growing body part. In addition to the two different
DNA extraction protocols were used in this study to compare the quality and
quantity of DNA yielded using different body parts. According to our study using
Phenol Chloroform method using without liquid Nitrogen yielded DNA with high
purity. Phenol Chloroform method using with liquid Nitrogen yielded high
concentrations of DNA.
